Shape determination: applications to proteins

Small-angle X-ray scattering methods have gained prominence as a rapid and complementary biophysical technique for shape determination of proteins. In this talk I will provide an overview of process, focusing technical details affecting sample preparation and data collection that leads to a successful shape determination of proteins. Technical considerations, including pitfalls, will be discussed. The utility of shape determination in addressing biological questions will also be illustrated using examples from literature and my own laboratory, particularly in the context of complementary data obtained from other biochemical and/or biophysical techniques.
