Next to E.coli expression the insect cell baculovirus expression system is the most widely used system for producing recombinant proteins for research applications, in particular proteins destined for structural analysis. Kinases, nuclear receptors, proteases and many other types of enzymes have been successfully produced. The application of baculoviruses for protein production in mammalian cells also exists due to the recent observation that modified baculoviruses containing mammalian cell-active expression cassettes can be used for gene delivery into mammalian cells. This presentation will focus on the attributes of the baculovirus system that have led to its successful application for producing proteins for structural analysis.

A number of examples will be provided and important considerations for its application in high-throughput expression and scale-up will be discussed.
