M. Roessle - Data Recording, Reduction and Processing

Data Recording, Reduction and Processing

Manfred Roessle
EMBL, Hamburg Outstation, Germany

This lecture will explain the initial steps of a solution SAXS experiments. After an introductory part of SAXS instrumentation, the different data reduction and processing steps will be discussed in detail:

  1. Recording of Small Angle Scattering (SAS) data
    1. SAXS beamlines;
    2. SAXS beamline optics;
    3. detection of X-rays for SAXS;
    4. automation.
  2. Primary Data Reduction
    1. radial averaging;
    2. normalization;
    3. subtraction;
    4. pipeline for data analysis.
  3. Initial data processing
    1. Guinier approximation;
    2. estimation of the molecular weight;
    3. quality check of SAXS data.
